On chromosome 5, the credible set includes links to LINC00461 and TMEM161B (Supplementary Data 3). The function of LINC00461 is unclear, but the RNA has highly localized expression in the brain81 and the genome-wide significant locus overlaps with variants in LINC00461 associated with educational attainment78. Alternatively, a genome-wide significant SNP in this locus (rs304132) is located in MEF2C-AS1, of strong interest given previous associations between MEF2C and severe intellectual disability,82–84 cerebral malformation83, depression70, schizophrenia33 and Alzheimer’s disease85, but the corresponding variant is not supported by the credible set analysis. Credible set annotations for other significant loci are similarly cryptic.
