We estimated heritability of PTSD in the EUA studies (Table 1) based on information captured by genotyping arrays (h2SNP) from summary statistics using LDSC12. Assuming a population prevalence of 30% after trauma exposure, overall h2SNP in PGC2 was 0.05 on the liability scale (P = 3.18 × 10−8). However, female heritability was highly significant (h2SNP = 0.10, P = 8.03 × 10−11), while male heritability was not significantly different from zero (h2SNP = 0.01, P = 0.63).Table 1Heritability estimates in subjects of European ancestry (EUA)Sample N N 10% prev30% prev50% prevCasesControls h 2 SNP 95% CI h 2 SNP 95% CI h 2 SNP 95% CIp-value All PGC223,212151,4470.040.02–0.050.050.03–0.070.060.04–0.083.2 × 10−8 PGC1.512,82335,6480.030.01–0.060.050.01–0.080.050.01–0.090.011 Men UKB10,389115,7990.130.1-0.150.170.14–0.210.190.15–0.232.1 × 10−18 PGC2990875,6050.01−0.02 to 0.030.01−0.03 to 0.050.01−0.03 to 0.050.63 PGC1.5636423,9050.01−0.04 to 0.050.01−0.05 to 0.070.01−0.05 to 0.080.69 UKB354451,7000.110.04–0.170.150.05–0.240.160.05–0.261.4 × 10−3 Women PGC212,97373,6270.070.05–0.090.100.07–0.130.110.07–0.148.0 × 10−11 PGC1.5612895280.150.08–0.220.210.11–0.310.230.12–0.332.7 × 10−5 UKB684564,0990.140.1–0.180.190.13–0.250.210.14–0.272.0 × 10−10Estimates are calculated using LD-score regression (LDSC) at different population prevalences after trauma exposure for the combined PGC freeze 2 samples, and separately for PGC1.5 (without the UK biobank), the UK biobank, and for men and women.
