Differences in genetic correlations between posttraumatic stress disorder and alcohol-related problems phenotypes compared to alcohol consumption-related phenotypes.

PTSD commonly co-occurs with increased alcohol consumption and AUD. While our previous work has demonstrated that there is a molecular genetic correlation between PTSD and alcohol-related problems (Sheerin et al., 2020), the goal of this study was to determine whether the genetic correlations with PTSD extend to alcohol consumption, and if the architecture of the genetic association with PTSD differs for alcohol consumption and alcohol-related problems. Further, we aimed to test these associations using both EA and AA summary statistics using data from the latest GWASs of PTSD, alcohol consumption-related, and alcohol-related problems. Among EA analyses, this study found positive and significant genetic correlations between PTSD and alcohol-related problems phenotypes, whereas negative with non-significant genetic correlations observed for PTSD and alcohol consumption-related phenotypes. These results indicate that which alcohol phenotype one uses in analyses absolutely matters, and that alcohol use is certainly not ‘close enough’ as a proxy for alcohol-related problems in examining its genetic associations with other conditions.
