Mice were then tested for their ability to ‘unlearn’ the previous location of the platform and to learn its new location. The location of the hidden platform was moved to the opposite quadrant of the water maze (Quadrant 3), and mice were tested for acquisition of the new location. After reaching the 15-second group-wide criterion, mice underwent a second probe trial to assess quadrant preference after reversal. During the reversal probe trial, normal C57/BL6 mice were shown to spend more time in the most recent quadrant (Q3), than in the initial learning quadrant (Q1) (Moy et al., 2007). Thus, mice with perseverative reversal learning deficits were expected to spend more time in the former quadrant (Q1) than unimpaired mice.
