Interleukin-6 (IL6) binds a cytokine receptor and triggers a signal transduction cascade through Janus kinases (JAK) that culminates in phosphorylation of STAT3 on the Tyr705 (Kaptein et al., 1996; Stark and Darnell, 2012). There is a strong correlation between high levels of the phosphorylated STAT3 (STAT3_pY705) and enrichment of the IL6 JAK STAT3 hallmark (IC = 0.422, p = 2.38×104).
